NUCLEIC ACID ANALYSIS

HIGH THROUGHPUT NUCLEIC ACIDS USING QIASYMHONY ROBOT FROM DIFFERENT MATERIALS

  • blood
  • tissues
  • cell cultures
  • saliva
  • tumors
  • bacteria
  • viruses (nucleic acids isolation from cultures, blood)
  • biotraces (min. 96 samples) from cheek epithelium swabs, blotting paper (blood from newborn screening) and other biotraces

Please note that plasmid isolation (mini, midi, maxiprep) is possible only manually.

GENOTYPING OF SINGLE KNOWN MUTATIONS-DIFFERENT TECHNIQUES

  • ARMS-PCR (amplification refractory mutation system)
  • RFLP (restriction fragment length polymorphism)
  • HA (heteroduplex analysis) using capillary electrophoresis, possible in about 80% of the mutations
  • HRM (high-resolution melting curve) using Qiagen Rotor Gene Q or Roche LightCycler

All of these techniques require optimization. For this purpose, it is required to provide a positive control verified by direct sequencing (Sanger or Pyrosequencing).

GENOTYPING OF SINGLE UNKNOW MUTATIONS- DIFFERENT TECHNIQUES

  • Pirosequencing – high sensitivity sequencing to find specific mutations (length of analysed sequence ~80 bp) , using Qiagen PyroMark Q24
  • NGS (next-generation sequencing) – simultaneous sequencing of multiple amplicons in many assays in a single tube, using Illumina MiSeq
